Abstract Molecularly targeted precision treatments have significant potential to improve therapy options for hard-to-treat cancers and reduce late effects in general. Genome sequencing has laid the foundation for precision medicine, yet, clinical success remains moderate. Identifying therapeutic targets at the protein and pathway level holds great promise, but may be considered complex. This is particularly true for smaller centres without local proteomics expertise, raising concern of new access barriers and increased inequity. Here, we present a road map for the nation-wide integration of pre-clinical proteomics evidence into pediatric precision oncology and provide proof of concept for proteome guided therapies.
