Abstract

Validation was done using SOP-007.01 based on the method of the DNeasy PowerSoil Pro kit (Qiagen, Hilden, Germany). Samples used for eDNA extraction were collected during the International and the German Small Scale Bottom Trawl Surveys (IBTS & GSBTS) implemented in research cruise WH428 in 2019. All samples were extracted by each conductor, i.e. A-samples by conductor 1, B-samples by conductor 2, and applying the four-eyes-principle. DNA yields and purity were photometrically measured and downstream performance was additionally tested with COI and 12S rRNA PCR using 1:5 diluted DNA extracts as templates. All validation experiments are deposited as e-protocols (GEN088 - GEN095).
