A Cu(OTf) 2 ‐promoted one‐pot three‐component reaction using rongalite as a C1 synthon for the synthesis of pyrimidine‐fused quinoline compounds has been reported. This protocol is characterized by readily available chemicals and straightforward operation, delivering the products in moderate to good yields (32 examples, 21%–86% yields). This strategy is suitable for large‐scale preparation and late‐stage derivatizations. The product with simple modifications exhibits significant aggregation‐induced emission properties and viscosity‐responsive fluorescence behavior.
