Jasplakinolide-like natural products and their analogs are F-actin-stabilizing cyclic pep-tides which are widely used in cell biology as tools for visualization and modulation of actin-dependent processes. In this study, we report the synthesis of a focused library of different compounds using an optimized synthetic strategy. Comprehensive in vitro and in cellulo evaluation facilitated a deeper understanding of structure-activity relationships (SAR) and resolved conflicting data concerning jasplakinolide H.
