Drought stress is a major factor in reducing the growth, development and production of plants. Therefore, studying the plants during the drought stress is highly useful to discover the new gained characteristics of plants. The potato is considered one of the most important economical crops in Egypt and over whole world. The main target is proline rich protein that will be extracting from potato plant under drought stress.Stress experiment was applied to two potatoes (Solanum tuberosum L.) cultivars Desiree and Diamante. The stress experiment was conducted by `polyethylene glycol (PEG) 4000, and water potentials were: zero (control), (PEG 2%), (PEG 4%) and (PEG 8%), after drought stress, the solutes accumulation in two potatoes cultivars were determined. The results showed that increasing the concentration of PEG resulted in a reduction of the percentage of plantlets survival, and reduced plantlets height, and plantlet dry weight but our previous studies revealed that the content of proline increased at higher PEG treatment. Moreover, the proline gene was isolated partially from the potatoes (Solanum tuberosum L). The alignment showed high similarity with other predicted corresponding from other cultivars.
