The conventional paraffin method is unsuitable for root specimens of Thismia (Thismiaceae), a genus of fully mycoheterotrophic plants, since it causes root tissues and associated fungal structures to become distorted or collapsed. Here, we present a modified paraffin method protocol optimized for vulnerable Thismia roots that mitigates this issue, with comparisons to the conventional method.
