Introduction: Azelnidipine (AZL) and Olmesartan (OLM) have emerged as effective antihypertensive agents, both as monotherapy and in combination. The newer combination of AZL and OLM improves hypertension. The combination was created to improve stage II hypertension treatment. AZL is a calcium channel blocker, and OLM is an angiotensin II type-1 receptor antagonist. This combination was developed to enhance treatment efficacy for stage II hypertension. This study provides a comprehensive and current overview of AZL and OLM analysis methods. It covers spectroscopic, chromatographic, and other analytical methods in detail. Methods: Relevant publications on AZL and OLM, focusing on estimation, analytical method development, and pharmaceutical formulations, were rigorously retrieved from databases such as MEDLINE, Scopus, and Web of Science using relevant keywords. Results: The methods available for AZL and OLM include spectroscopy, hyphenated techniques, high-performance liquid chromatography, and thin-layer chromatography. All reported methods are recognized for being simple, accurate, economical, precise, and reproducible. Discussion: Analytical method development enables accurate quantification of Azelnidipine and Olmesartan in combination therapies, ensuring efficacy, safety, and regulatory compliance through robust RP-HPLC, UPLC-MS, and LC techniques aligned with Quality by Design principles. Conclusion: This review offers a comprehensive collection of various analytical techniques published for AZL and OLM, both individually and in combination, serving as a valuable resource for researchers in their future work. Recent advancements in analytical methods have enabled the development of sensitive, selective, and reliable methods for quantifying drugs in various matrices.
