Colorectal cancer (CRC) is a digestive tract tumor and ranks as the third most common malignant tumor globally. This study aimed to confirm the effects of miR-3928/IQGAP3 on the development and prognosis of CRC. We assessed the prognostic value of miR-3928 through survival analyses using Kaplan-Meier plots and Cox regression modeling. The interaction between miR-3928 and IQGAP3 was confirmed by RT-qPCR and Dual-Luciferase assay. We used CCK8, Transwell assay, and colony formation assay to examine their effects on SW480 and HT29 cell proliferation and migration. Low miR-3928 expression correlates with poor prognosis in CRC patients, while IQGAP3 is highly expressed in CRC. MiR-3928 targets and regulates IQGAP3. Overexpression of miR-3928 inhibits the proliferation and migration of CRC cells, but overexpression of IQGAP3 reverses this effect. Additionally, miR-3928 overexpression promotes ferroptosis, which may be a key mechanism underlying its ability to inhibit CRC cell proliferation. In CRC, low miR-3928 expression correlates with poor prognosis. MiR-3928 suppresses CRC cell proliferation and migration by targeting IQGAP3, possibly through ferroptosis promotion. This study confirms that miR-3928 acts as a tumor suppressor in CRC, offering a potential target for prognostic evaluation.
