Background: Despite advances in frontline therapy, most patients (pts) with adv/met NSCLC experience progression, and later-line chemotherapy with docetaxel-based regimens has limited benefit. Dato-DXd is an antibody-drug conjugate composed of a TROP2 directed monoclonal antibody covalently linked to a highly potent cytotoxic payload via a stable, tumor-selective, tetrapeptide-based cleavable linker. This is the first report of results from TROPION-Lung01 (NCT04656652), a randomized, global, open-label, phase 3 study comparing the efficacy and safety of Dato-DXd vs docetaxel in pts with adv/met NSCLC with or without actionable genomic alterations (AGAs).
