We report a simplified immunosensing system for determining Interleukin-1α (IL-1α) levels in the stratum corneum. Our system measures IL-1α as a color change of 3,3’,5,5’-tetramethylbenzidine and estimates the concentration from an optimized calibration curve derived from the weighted RGB information. We tested the IL-1α measurement system using tape-stripped skin samples, and it showed a strong correlation (r=0.896) with conventional methods. Additionally, normalizing the IL-1α level based on total protein allowed appropriate comparisons between samples collected from different subjects. The results indicated that our system could support routine skin monitoring for beauty purposes.
