High Resolution Image Download MS PowerPoint Slide Quantitative assessment of renal function decline in early stages is crucial for effective management of kidney diseases. Herein, leveraging the high spatiotemporal resolution and deep tissue penetration depth of computed tomography (CT) technology, we achieved early and quantitative detection of renal dysfunction of bilateral and unilateral kidney diseases using Au 25 SG 18, a representative renal-clearable gold nanoparticles with atomically precise structure, high renal clearance efficiency, minimal extrarenal accumulation and good biocompatibility. By monitoring the intrarenal transport kinetics of Au 25 SG 18 via CT imaging, we quantified its intrarenal transport rate (ITR) and dynamically visualized its intrarenal distribution across the parenchyma and pelvis. Under kidney diseases of cisplatin-induced bilateral injury and ureter-obstructed unilateral injury, both the ITR and spatial intrarenal distribution of Au 25 SG 18 were both sensitively altered with 28% and 74% of ITR reduction in bilateral and unilateral injuries, respectively. These indicators also realized earlier reporting renal dysfunction than conventional blood biomarkers. Our work not only established a method for quantifying the intrarenal transport of renal-clearable gold nanoparticles, but also introduced a class of CT contrast nanoagent for quantitative evaluation of renal function loss.
