Abstract The combination of DPNH with liver alcohol dehydrogenase (alcohol-NAD oxidoreductase, EC 1.1.1.1) has been examined by rapid spectrofluorimetry. The time course of the fluorescence was the same using six combinations of exciting and observing wave lengths, and could be accounted for by the scheme E + R ⇌ ER where E is the free enzyme and R is DPNH. The value of the combination velocity constant was 5 x 106 m-1 sec-1 at 3°. There was no evidence for the formation of more than one enzyme-DPNH complex.
