The function of neutrophils in the tumor microenvironment is not fully understood, in part because of technical challenges posed by their short half-life that limit single-cell profiling opportunities. Wu et al developed a neutrophil-sorting protocol to enable single-cell RNA-sequencing analysis of matched tumor and normal tissue samples and identified 10 distinct neutrophil clusters. HLA-DR+CD74+ antigen-presenting neutrophils are among the most preferentially enriched in tumors, and metabolomic and epigenetic analyses indicate that leucine-activated neutrophil antigen presentation activates antitumor T cells and enhances the antitumor efficacy of anti–PD-1 in mouse models, highlighting a potential therapeutic opportunity.
