Abstract Approximately 50% of pancreatic cancer patients are diagnosed as stage IV with poor prognosis and show treatment resistance to conventional cytotoxic chemotherapy and recently developed immunotherapy. Therefore, there is an urgent need in developing novel treatment options for pancreatic cancer patients and examine them by using proper preclinical models. Murine orthotopic tumor model is generated by seeding murine cancer cell lines or patient-derived cancer cells into the relevant organ of tumor origin. Significance of the orthotopic pancreatic cancer model have been highlited since it can be easily generated compared to GEMM(Genetically engineered mouse models) and represent more clinical relevance than subcutaneous model. Here, we described how to create the murine orthotopic pancreatic cancer model.
