Tetraphenylene (TPE) and its derivatives are highly studied materials in the field of aggregation-induced electrochemiluminescence (AIECL). We successfully synthesized a block-like metal–organic framework by employing Zn2+ and the TPE-based ligand H8ETTB (H8ETTB = 4′,4‴,4‴″,4‴‴′-(ethene-1,1,2,2-tetrayl) tetrakis ((1,1′-biphenyl-3,5-dicarboxylic acid))), which demonstrates excellent ECL properties. For simplicity, we refer to this material as the Zinc-based metal–organic framework (Zn-MOF). H8ETTB is a ligand and luminophore, increasing the loading of the luminophore. The Zn-MOF exhibits considerable specific surface area and porous structure, which enables efficient reaction between the luminophores and coreactants, resulting in excellent ECL. The ECL emission of Zn-MOF is more intense than that of the H8ETTB monomer and aggregates. Due to the distinctive ECL characteristics of Zn-MOF, we employed it as an ECL luminophore in an ECL immunoassay, resulting in the establishment of an ECL system for the extremely sensitive detection of 5-fluorouracil. PdAu nanoflowers (PdAu NFs) were utilized as electrodes, which were modified because of their considerable specific surface area and electrocatalytic efficiency. As a result, they additionally improved the ECL performance. The design of the immunosensor is relatively straightforward, facilitating their manufacture. Furthermore, the immunosensor was successfully tested in human serum samples, demonstrating comparable detection results to those obtained using high-performance liquid chromatography with tandem mass spectrometry (HPLC-MS/MS). This research thus offers a practical method to enhance the performance of ECL and introduces an approach for identifying 5-FU in clinical analysis.
