Abstract Introduction: Ayurveda, the ancient Indian system of medicine, emphasizes the use of natural formulations to restore balance and health by addressing the root causes of diseases. In the global resurgence of interest in traditional medicine, Ayurvedic polyherbal formulations have gained wide recognition for their safety and efficacy. Standardization of these formulations is essential to ensure reproducibility, therapeutic consistency, and global acceptance. This study focuses on the standardization and chemical profiling of Bhringarajaadi Taila to validate its pharmaceutical properties. Objectives: To standardize the preparation of Bhringarajaadi Taila , a classical Ayurvedic medicated oil, and conduct comprehensive analytical evaluations to assess its physicochemical and phytochemical characteristics in alignment with Ayurvedic Pharmacopoeia standards. Materials and Methods: Bhringarajaadi Taila was prepared as per the classical reference from Yogaratnakara, using ingredients including Bhringaraja , Triphala , Sariva , Mandoora and Tila Taila as the base. Organoleptic and physicochemical parameters such as moisture content, viscosity, and refractive index were evaluated. High-performance thin-layer chromatography (HPTLC) was conducted to identify and validate marker compounds, including Wedelolactone from Bhringaraja and Sesamin from sesame oil. Retention factor (Rf) values were recorded under different solvent systems to develop specific chemical fingerprints. Results: The oil exhibited acceptable organoleptic qualities and physicochemical properties consistent with Ayurvedic Pharmacopeia standards. Moisture content was within the permissible range, viscosity and refractive index values indicated proper consistency and purity. HPTLC analysis revealed distinct Rf values corresponding to Wedelolactone and Sesamin, confirming the presence of key phytoconstituents. The fingerprinting validated the formulation's consistency and authenticity. Conclusion: The standardized preparation of Bhringarajaadi Taila was found to comply with classical Ayurvedic methods and modern pharmaceutical parameters. Its chemical profile and therapeutic potential support its traditional use in managing scalp conditions.
