Abstract The transcription factor PAX8 has become one of the most promising selective targets for high grade serous ovarian cancer. However, mechanistic insight into PAX8's interactions and how this knowledge can be applied toward a therapeutic approach remains elusive. Here we investigate novel mechanistic roles of PAX8 in ovarian cancer. We identified direct interactions between PAX8 and other transcriptional coregulators and characterized how these interactions occur. Additionally, we are conducting PAX8 resistance CRISPR screens to identify new targets that suppress proliferation defects associated with PAX8 knockout. Together, our findings reveal important insight into PAX8’s tumorigenic roles and how it can be targeted therapeutically. Citation Format: Anna Russo, Kostianna Sereti, Ryan Raisner, Karen Gascoigne.
