Abstract Description Advancements in flow cytometry and fluorescent dyes have enabled the development of more complex and detailed panels, allowing for the detection of a broader range of markers and cell sub-populations, even in limited samples. However, these larger panels also present challenges in design, optimization, and data analysis. In this context, we have established a procedure to test a multicolor panel using the CytoFLEX mosaic Spectral Detection Module, the latest addition to our CytoFLEX family for spectral analysis. Our comprehensive flow cytometry panel is designed for immunophenotyping various immune cell subsets, including B cells, T cells, and myeloid cells. The panel targets lineage markers to identify and characterize a diverse range of immune cells, such as CD4 and CD8 T cells, regulatory T cells, γδ T cells, NKT-like cells, B cells, NK cells, monocytes, and dendritic cells. This panel enables a detailed analysis of the functional states of these immune cells, which is particularly valuable in research settings with limited sample availability. By maximizing the amount of information obtained from a single sample, our approach enhances the efficiency and effectiveness of immune cell analysis in such settings.
