Abstract

Dual PD-1/CTLA-4 inhibition improves outcomes in several cancers but remains limited by toxicity. A recent article reports the first-in-human study of volrustomig, a novel PD-1/CTLA-4 bispecific antibody, demonstrating durable responses, but dose-dependent immune-related adverse events were also observed. This commentary examines whether the bispecific format meaningfully improves the therapeutic index and considers priorities for the next phase of development.
