Abstract Grapevines acclimate to water stress primarily through a combination of stress-avoidance and stress-tolerance mechanisms. From this perspective, we revisit the potential interactions between grapevine scions and rootstocks under drought, emphasizing their specificities across a broad biological space spanning both intra- and inter- Vitis diversity as well as their associated microbiomes. Building on the described ecophysiological responses, we summarize the main known biomolecular adjustments and their potential applications in assisted breeding programmes.
