Doxorubicin (DOX) is extensively used as an antitumor drug, but its severe cytotoxicity limits its therapeutic use. This study was designed to explore the potential protective impact of watercress extract (WCE) at a dose of 500mg/kg/day against hepatotoxicity induced by DOX (2mg/kg, twice/week) in rats, in two-month experiment. Injection of DOX significantly elevated serum levels of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP) and total bilirubin (TB), along with markedly reduced serum contents of total proteins (TP) and albumin (ALB). Parallelly, DOX produced hepatic oxidative stress by increasing concentrations of malondialdehyde (MDA), protein carbonyl (PC) and nitric oxide (NO), and by lowering the levels of antioxidants including glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GRd) and glutathione-s-transferase (GST). Furthermore, DOX induced marked increases in serum levels of interleukin-1 beta (IL-1β), interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α) and intercellular adhesion molecule 1 (ICAM-1). In addition, DOX produced apoptosis through increasing p53 and caspase-3 levels, along with decreasing Bcl-2 content. Histological examination confirmed these biochemical findings, revealing obvious hepatic damage. Administration of WCE effectively attenuated DOX-induced oxidative stress, inflammation, apoptosis and histopathological changes. In conclusion, obtained results evidenced a novel beneficial role of WCE in mitigating DOX-induced hepatotoxicity.
