Purpose: Plant-derived exosomes are promising bioactive mediators of intercellular communication. This study aimed to isolate and characterize exosomes derived from Eclipta prostrata and evaluate their potential anti-inflammatory and moisturizing effects on the skin. Methods: E. prostrata exosomes were isolated using the ExoQuick system and characterized through nanoparticle tracking analysis and an antibody array. Their biological activities were evaluated. Results: E. prostrata-derived exosomes had an average diameter of 107±5.4 nm and a concentration of 1.82×1011 particles/mL. Treatment with E. prostrata-derived exosomes significantly inhibited nitric oxide production and downregulated COX-2 protein expression in LPS-stimulated RAW 264.7 cells. In keratinocytes, there was a concentration-dependent increase in hyaluronic acid production compared to the positive control, indicating enhanced skin hydration potential. Conclusion: These results suggest that E. prostrata-derived exosomes possess physiological activities and could serve as promising natural functional ingredients for cosmeceutical applications.
