Abstract BACKGROUND: Triphalaguduchyadi Vati (TGV) is a poly herbal formulation based on one of the formulations indicated in Sthaulya (obesity) in Astanga hridaya . This formulation is a blend of five drugs, and quality analysis is essential before conducting its therapeutic evaluation. Hence, this study aimed to evaluate TGV through pharmacognostical, physico-chemical parameters and HPTLC fingerprinting in the process of establishing quality control (QC) data. METHODS: Pre-authenticated raw drugs were procured and TGV was prepared at the pharmacy, ITRA, Jamnagar. Organoleptic parameters and microscopic analysis of Vati were done at pharmacognosy department, ITRA, Jamnagar. Physio-chemical analysis; Heavy metal analysis and HPTLC of TGV were done at the pharmaceutical laboratory, ITRA (Jamnagar) and Vasu Research Center (Vadodara), respectively. RESULT: Microscopic features of TGV were cross-checked with API standards of individual ingredients and it was found to have characteristics of all ingredients in it. In pharmaceutical parameters; pH value was 7.0, the water soluble extractive value was 12.7% w/w, the methanol soluble extractive value was 0.8% w/w, the ash value was 8.19% w/w, the loss on drying was 5.95% w/w. Heavy metals were under the permissible limit of API. HPTLC at 254 nm, 366 nm, and 540 nm showed 7, 7, and 9 spots respectively. CONCLUSION: This study has generated preliminary data on Pharmacognostical, pharmaceutical parameters, and HPTLC. This fingerprinting can be useful for future researchers to reproduce this formulation and to establish the QC parameter of TGV.
