The Qizhen Shengbai compound (QZSBC) is utilized for the adjunctive treatment and prevention of radiation-induced damage to the hematopoietic system. This study aims to optimize the extraction process of QZSBC. Through network pharmacology prediction and high-performance liquid chromatography (HPLC) verification, along with reference to the 2020 edition of the "Chinese Pharmacopoeia," the key components of QZSBC were identified as salidroside, echinacoside, specnuezhenide, and betaine. The pharmaceutical research was conducted on samples from three extraction processes to detect the content of index components. A comprehensive comparison revealed that the total content of index components was highest in the water-extracted samples. The pharmacodynamic test results showed that the water extraction group had the best effect on the recovery and improvement of hematopoietic system injury of radiation mice. Through orthogonal experiments, it was established that the optimal extraction process involves adding 12 times the volume of water and conducting three rounds of reflux extraction, each lasting 1 h. This study is the first to determine the extraction process of QZSBC through the combination of multiple methods and optimize the extraction process through orthogonal experiments, providing research ideas for the subsequent development of QZSBC.
