The COVID-19 pandemic has rapidly spread across global societies, caused by the SARS-CoV-2, leading to significant challenges and impacts. The development of antiviral drugs targeting pivotal viral proteins, particularly the S-protein, Mpro, PLpro, and RdRp, offers a promising strategy for addressing the complexity of treating COVID-19. There is a need to focus on the development of COVID-19 drugs that aim at multiple targets. In particular, developing effective antiviral drugs is essential for addressing potential future pandemic threats. Hence, this review highlights the principal COVID-19 targets and other possible targets, and offers critical insights into the ongoing progress toward developing effective treatments for COVID-19 and other emerging coronaviruses.
