Introduction: The topical administration of medicinal substances has demonstrated encouraging results in managing skin conditions. Skin conditions are among the conditions that have been treated with siRNA-based therapeutics. Treatment options for skin conditions have expanded due to the topical application of siRNA-based medicines. The quick breakdown and low cellular absorption of siRNA restrict its application. Furthermore, a key obstacle to the administration of topical medicines is the top layer of skin known as the stratum corneum. Methods: An effective topical formulation is needed to overcome the inherent limitations of siRNA delivery and distribute the siRNA to the site of action. Non-viral techniques and active penetration techniques have been used together to deliver siRNA topically for the mitigation of skin diseases. Results: Preclinical testing has been done on several delivery strategies, and in a few instances, the outcomes have indicated potential for clinical trials. The utilization of multiple delivery strategies for effective distribution at the targeted site and updates on developments in non-viral siRNA delivery for skin disorders are covered in this review. Conclusion: Genetic engineers and gene therapists are actively advancing their strategic approaches to vanquish the challenges. They are refining vectors to maximize efficiency and exploring innovative carriers that have latent prospects for perfect gene delivery. As this dynamic field continues to evolve, the collaborative efforts of technical professionals will be pivotal in shaping the future of gene therapy.
