Abstract

Abstract The total synthesis of natural products, especially those derived from marine sources, continues to advance the field of synthetic chemistry. Norfuranocembranoids, a class of diterpenoids originating from marine organisms like soft corals, have attracted significant attention due to their unique structures and promising biological activities, including anti‐inflammatory and antifungal properties. Among these compounds, ineleganolide, sinlochmodin, scabrolide, and horiolide are noteworthy targets due to their complex ring systems and stereochemical intricacies. The synthesis of polycyclic compounds involves key reactions such as the Luche reduction for selective diastereoselective reductions and Ueno–Stork radical cyclization for precise stereo‐control. Strategies like Mukaiyama–Michael addition, Appel reaction, and Stork–Zhao olefination facilitate carbon‐carbon bond formation. Additionally, transformations using Burgess reagent and retro‐aldol reactions are crucial for achieving the desired stereochemistry. These methods help create the polycyclic structures of norfuranocembranoids, allowing for the exploration of their potential biological benefits. This review aims to concisely compile all the syntheses in one place so that these compounds can be synthesized and utilized for their various activities.
