Abstract

溶酶体(Lysosome)是负责细胞内物质降解与稳态调控的重要细胞器,它与内体、自噬体、线粒体等协同工作,承担胞内蛋白质、糖类、脂类及核酸等大分子的降解与循环利用。溶酶体功能受损诱发并加重溶酶体贮积病、神经退行性疾病、和代谢疾病等。但促进溶酶体生成并增强溶酶体降解功能可以清除治病底物,进而改善相关疾病。ZKSCAN3(Zinc Finger With KRAB And SCAN Domains 3;ZNF306)是调控溶酶体生成的重要蛋白,做为含有SCAN与KRAB结构域的锌指转录抑制因子,在转录水平上调控自噬与溶酶体相关基因的表达。ZKSCAN3亚细胞定位、生物活性、和蛋白水平的变化(二聚体和翻译后修饰)影响靶基因的转录调控,进而影响生理条件下的稳态调控和病理条件下的疾病发生发展。
