Identification, biological characteristics, and screening of fungicides for the pathogen causing lotus leaf marginal necrosis

Abstract

【目的】为明确莲叶缘病的病原菌种类,探究生物学特性及有效防治药剂,为该病害的防治提供依据。【方法】本研究采用组织分离法对采自江西省广昌县的莲叶缘枯病病叶进行病原菌分离纯化,并通过柯赫氏法则验证其致病性。综合形态学和多基因序列(ITS、TUB2和TEF1-α)分析鉴定病原菌的种类,以菌丝生长速率法测定其生物学特性,并测定8种常见杀菌剂对病原菌菌丝生长的抑制作用。【结果】结果表明,从采集的病叶组织中分离出的4株病原菌,在PDA培养基上培养特征一致,选取其中一个菌株LB1并结合形态学和分子生物学鉴定,确定造成莲叶缘病的病原菌为橘色黑孢霉(Nigrospora aurantiaca)。该病原菌菌丝生长最适温度为25℃,最适pH为6,最适培养基为马铃薯葡萄糖培养基(PDA),最适碳、氮源分别为蔗糖和硝酸钾。8种药剂室内毒力测定结果表明,450 g/L咪鲜胺EW、300 g/L苯甲·丙环唑EC对菌丝生长抑制效果较好,EC50分别为0.022 mg/L、0.648 mg/L。【结论】本研究首次报道橘色黑孢霉(N. aurantiaca)引起莲叶缘病,该病害田间防治首选450 g/L咪鲜胺EW,300 g/L苯甲·丙环唑EC可作为轮换用药选择。
