Key Points

  • This research aims to evaluate the antimicrobial properties and safety of novel phosphonium derivatives.
  • Synthesis of phosphonium derivatives PP8 and 6.5
  • Assessment of membrane depolarization and permeabilization effects
  • Evaluation of biofilm formation prevention and disruption
  • Investigation of mutagenic properties using the Ames test
  • PP8 and 6.5 caused depolarization and permeabilization of bacterial membranes.
  • Both compounds effectively prevented and disrupted biofilm formation in Staphylococcus aureus.
  • No resistance development was observed in the S. aureus strain.
  • Compound PP8 showed no mutagenic effects in the Ames test.

Abstract

New derivatives of phosphonium salts (Z)-(2-(2-hydroxy-5-chlorphenyl)-2-phenylethenyl)octyldiphenylphosphonium chloride (PP8) and (2-hydroxybenzyl)dodecyldiphenylphosphonium chloride (6.5) were found to cause depolarization and permeabilization of the bacterial membrane. The ability of phosphonium salts to prevent the formation of Staphylococcus aureus biofilms and to disrupt them was demonstrated. Furthermore, the results demonstrated that the substances do not cause resistance development in the S. aureus strain. Finally, compound PP8 did not demonstrate mutagenic properties in the Ames test using strains of Salmonella typhimurium TA100 and S. typhimurium TA98.
