Abstract

Introduction:: The present study aimed to evaluate the diuretic potential of the ethanolic extract of Holoptelea integrifolia leaves, a plant traditionally used in Indian medicine, using Wistar albino rats as the experimental model. Methods:: The study was conducted on randomly selected Wistar albino rats. Before administration, the animals were fasted for 20 hours. Four groups were established: Group 1 (Negative Control) received normal saline; Group 2 and Group 3 received 100 mg/kg and 200 mg/kg of Holoptelea integrifolia ethanolic extract, respectively; Group 4 (Positive Control) received furosemide at 20 mg/kg. Diuretic activity was assessed by measuring urine volume and electrolyte excretion. Results:: Both 100 mg/kg and 200 mg/kg doses of Holoptelea integrifolia extract significantly increased water and salt excretion compared to the negative control. The diuretic index values were 2.57 and 2.81, while the diuretic activity values were 0.13 and 0.14 for the respective doses. Although these values were lower than those observed with furosemide, they demonstrated a clear diuretic effect. Discussion:: The findings confirm the diuretic efficacy of Holoptelea integrifolia ethanolic extract, supporting its ethnopharmacological use. The dose-dependent increase in diuretic parameters suggests the presence of bioactive compounds influencing renal function. However, the activity was less potent than the standard drug furosemide, indicating the need for further phytochemical and mechanistic studies. Limitations include the absence of long-term toxicity data and detailed electrolyte profiling. Conclusion:: This study substantiates the diuretic potential of Holoptelea integrifolia ethanolic leaf extract, aligning with its traditional medicinal applications. The results encourage further exploration into its active constituents and therapeutic viability in managing fluid retention disorders. conclusion: The diuretic index of ethanolic extract of Holoptelia integrifolia at 100mg/kg 200mg/kg is 2.51 2.81 when given to Wistar albino rat. The value of the diuretic index indicates that the Holoptelia integrifolia has good diuretic activity it produces dose-dependent action. (As dose increase, diuretic activity increases.)
