Abstract

MODULATION OF PULPAL MMP-8 AND VEGF-ALPHA BY BIOACTIVE CALCIUM SILICATE AND BLOOD COCKLE SHELL EXTRACT COMBINATION IN RATS Abstract Objectives: This study evaluated MMP-8 and VEGF-Alpha expression in rat dental pulp inflammation after application of bioactive calcium silicate combined with blood cockle shell extract. Methods: Forty mandibular incisors were randomly assigned to four groups: 100% bioactive calcium silicate (control) and three treatment groups containing 25%, 50%, or 75% shell extract with the remainder calcium silicate. Direct pulp capping was performed, and samples were collected on days 1, 3, 7, and 14. Results: Immunohistochemistry and one-way ANOVA revealed significant differences in MMP-8 expression on days 1 and 3, and VEGF-Alpha expression across all time points (p < 0.05). The 75% shell extract group showed the lowest MMP-8 and highest VEGF-Alpha expression. Conclusion: These findings suggest that combining blood cockle shell extract with bioactive calcium silicate enhances regenerative potential in inflamed pulp compared to only calcium silicate. Méthodes: Quarante incisives mandibulaires ont été réparties au hasard en quatre groupes : 100 % de silicate de calcium bioactif (témoin) et trois groupes de traitement contenant 25 %, 50 % ou 75 % d'extrait de coquille, le reste étant du silicate de calcium. Un recouvrement pulpaire direct a été réalisé et des échantillons ont été prélevés aux jours 1, 3, 7 et 14. Résultats: L'immunohistochimie et l'ANOVA à un facteur ont révélé des différences significatives dans l'expression de la MMP-8 aux jours 1 et 3, et dans l'expression du VEGF-alpha à tous les moments (p < 0,05). Le groupe contenant 75 % d'extrait de coquille a présenté l'expression la plus faible de MMP-8 et la plus élevée de VEGF-alpha. Conclusion: Ces résultats suggèrent que l'association d'un extrait de coquille de coque rouge et de silicate de calcium bioactif améliore le potentiel régénératif de la pulpe inflammatoire par rapport au silicate de calcium seul.
