Key Points

  • This research aims to enhance anti-tumor responses in glioblastoma using self-amplifying RNA delivered by nanoparticles.
  • Utilized poly(lactic-co-glycolic acid) nanoparticles to deliver IL-12 RNA.
  • Assessed interaction of nanoparticles with the tumor microenvironment.
  • Focused on macrophage recruitment and immune cell stimulation.
  • IL-12 delivery triggered significant cytokine release leading to extensive immune cell recruitment.
  • Amplified anti-tumor responses observed in the glioblastoma models post-treatment.
  • Nanoparticles effectively reshaped the tumor microenvironment, enhancing local immune activity.

Abstract

Self-Amplifying RNA Nanomedicine This cover illustrates Self-amplifying RNA-loaded Poly(lactic-co-glycolic acid) nanoparticles interacting with the tumor microenvironment. Upon reaching cancer cells, the nanoparticles release their RNA payload, triggering localized IL-12 expression and macrophage recruitment. The secreted cytokines recruit and stimulate immune cells, reshaping the tumor microenvironment and amplifying anti-tumor responses through a nanomedicine-based immunotherapy approach.
