Effect of an oxygen-releasing mouthwash on Aggregatibacter actinomycetemcomitans and HaCaT cells: A pilot study

Abstract

Abstract: Background: Oxygen-releasing oral care products are effective in treating ulcers, gingival disease, and root canal irrigation. The present in vitro study evaluated the antibacterial properties and biological effects of a commercially available oxygen-releasing mouthwash, Blue ® M. Materials and Methods: We compared Blue ® M mouthwash with chlorhexidine (CHX) using the agar disk diffusion assay and minimum inhibitory concentration methods. Furthermore, 1 μL/mL of mouthwash was applied to HaCaT cells to assess its effects on cell migration using the scratch migration assay. Results: The zone of inhibition for the two mouthwash formulations was comparable. Blue ® M mouthwash demonstrated the inhibition of Aggregatibacter actinomycetemcomitans (Aa) at 6.25 μg/mL, and 0.12% CHX was effective at 0.4 μg/mL. Blue ® M (1 μL/mL) promoted HaCaT cell migration by up to 88.5% at 72 h compared to the control, with no adverse morphological changes. Conclusions: Blue ® M was effective at inhibiting Aa and exhibited negligible cytotoxicity in HaCaT cells, with minimal effects on cell migration.
