Abstract

Replication of the simian virus 40 (SV40) genome in infected permissive cells begins at a unique site in the viral DNA, located at map coordinate 0.67 on the standard SV40 map, and proceeds bidirectionally around the circular molecule, terminating opposite the origin (Nathans and Danna 1972; Danna and Nathans 1972; Fareed et al. 1972). The origin of replication (ori) appears to be a specific nucleotide-sequence signal, whereas the terminus of replication coincides with the junction of the two growing forks rather than with a particular nucleotide sequence (Brockman et al. 1975; Lai and Nathans 1975). Analysis of deletion mutants and evolutionary variants of SV40 indicates that the ori sequence is within an 80-bp segment between map coordinates 0.66 and 0.675 (Lai and Nathans 1974; Shenk 1977; Gutai and Nathans 1978).
