Recently, ion transport phenomena in biological tissue environments have attracted considerable attention. We have also developed a novel method for measuring local proton transport near biological cells using triple-barreled glass microelectrodes. In this study, proton diffussion within layered gel structures are investigated by scanning and fixing the triple-barreled glass microelectrodes, where agarose gel sheets impregnated with pH buffer solutions are prepared and stacked to mimic the structure of biological tissues. Ions near the interface diffuse according to the concentration gradient when the separated agarose gel sheets of different pH values are contacted with each other. As a result, the time-dependent proton diffusion within the layered gel structures is evaluated and found to be in good agreement with theoretical analysis.
