Background: Saliva-based liquid biopsy offers non-invasive genotyping for OSCC. Methods: 33 pre-surgical saliva samples and matched tissue samples. Extracted salivary cell-free DNA; TP53 codon 72 polymorphism (c.215C>G) detected by ddPCR and compared with tissue DNA. Results: Genotypes: C/G 45.5%, G/G 36.3%, C/C 18.2%. Concordance observed between salivary cfDNA and tissue DNA. Conclusion: Saliva is a reliable source for TP53 codon 72 genotyping; supports feasibility of saliva liquid biopsy in OSCC.
