SUMMARY A serum-neutralization ( sn ) test for canine distemper virus ( cdv ) in cell culture in microtitration plates was found to be as sensitive as the conventional test in embryonating hen eggs. Serotest results of 148 samples indicated the microtest had either greater sensitivity or smaller measurement error. Reproducibility of serum titers using the microplate assay was analyzed. The combined effect of all sources of variability within a laboratory resulted in a standard error of ± 0.3 (log 10 ) for a sample of serum titrated at threefold dilutions with 4 replicates per dilution.
