Small molecule inhibits bacterial hsp70, disrupting protein homeostasis and indicating a new way to address antibiotic resistance in Gram-positive bacteria.
Towards the development of new strategies against pathogenic bacteria, we report a small molecule inhibitor of bacterial heat shock protein 70 (Hsp70), called DnaK, a central component of the bacterial protein chaperone network. Our pyridylamide-based peptidomimetic DnaK inhibitor disturbs protein homeostasis and impairs chaperone function, effectively disrupting Gram-positive bacterial growth and biofilm formation, offering a different approach to tackling antibiotic resistance.
