Analysis reveals increased GLUT-3 expression in childhood high-grade gliomas, suggesting key roles of these proteins.
AIMS Paediatric high-grade gliomas (pHGG) are the leading cause of childhood cancer deaths. A subset of pHGG has Histone 3 (H3) mutation in them, mainly H3-K27M, H3-G34R, H3-G34V mutations. Our previous research has found increased expression of cell membrane proteins, GLUT-1 (Glucose transport) and GLUT-3 in H3 mutated pHGG. The aim of this study is to analyse the role of GLUT-1 and GLUT-3 in H3 mutated pHGG by localization, expression and inhibiting their function these with drugs/small molecules. METHODS Expression and localization study of GLUT-1 and GLUT-3 proteins in different pHGG cultured cell-lines and controls, was carried out by immunofluorescence and western-blot experiments. Inhibition of GLUT-1 and GLUT-3 by Glutor measured by cell proliferation assays and analyses. RESULTS Immunofluorescence and western blot experiments revealed increased expression of GLUT-3 protein in pHGG cell lines than control astrocytes. Although over-expressed, GLUT-1 was less varied in expression than GLUT-3. Results of the above experiments and inhibitory effect of Glutor will be presented. CONCLUSION The study provides insight into the potential role of GLUT-1 and GLUT-3 proteins in H3 mutated pHGG and highlights areas for further research and could be targets for future therapeutics.
