This protocol details fixation and immunohistochemical staining for Aiptasia larvae, enabling clear microscopy imaging.
This protocol is for fixation of Aiptasia (Exaiptasia diaphana) larvae and subsequent staining of microtubules, filamentous actin and nuclei. The resulting samples can be imaged under a widefield fluorescence microscope with sufficient image quality to assess basic morphological features such as size, gastric cavity and pharynx, cilia and apical tuft. For highest quality images, a confocal laser-scanning microscope can be used.
