Type 2 Diabetes Mellitus (T2D) is a metabolic disorder caused by dysfunction insulin secretion by pancreatic beta cells. Blood isolation samples from the buffy coat, contained higher levels of DNA than whole blood samples. The aim of this research is to find out the differences centrifugation speed of blood samples to determine the concentration and purity of DNA in Diabetes examinationMolecular-based Type 2 Mellitus (T2D). The research was carried out quantitatively with descriptive research type experimentally with purposive sampling technique. The samples used were 5 blood samples from T2D patients which were divided into 5 treatment groups (without centrifugation, centrifugation speed 500 rpm, 1500 rpm, 3000 rpm, and 4500 rpm for 5 minutes). Test examinations are carried out qualitatively (electrophoresis) and quantitatively (Uv-Vis Spectrophotometer). Research data was analyzed using the one way Annova test. No there were significant differences in treatment.
