BACKGROUND MicroRNAs (miRNAs) are short noncoding RNAs with broad regulatory functions in cells. Among these, miR10a and miR10b (miR10a/b) have been widely studied in cancer research due to their expressional association with various malignancies. OBJECTIVE This study aims to investigate the expressional changes of miR10a/b in HL-60 and MOLT-4 human cell lines, which serve as model systems for acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL), respectively, and compare these levels with those in peripheral blood mononuclear cells (PBMCs). METHODS MiRNAs were selected through a multistep filtering method using various databases. Specific primers with complementary sequences for stem-loop structured RNAs were designed. RNAs were extracted from cultured cells, and their expressional levels were measured using the RT-PCR technique. RESULTS Data analysis revealed that the expressions of miR10a/b were increased in both cell lines, HL-60 and MOLT-4. CONCLUSIONS This result suggests that miR10a/b can serve as prospective biomarkers for prognostic and diagnostic targets in AML and ALL malignancies.
