Advanced Insulin Synthesis by One‐Pot/Stepwise Disulfide Bond Formation Enabled by S‐Protected Cysteine Sulfoxide

S-Protected cysteine and its sulfoxide pairs allow stepwise disulfide bond formation, with the reaction increasing in acidity. This system enables an unprecedented synthesis of insulin possessing three disulfides and its lipidated analog. Stepwise formation of three disulfides generally requires three independent oxidation steps; however, the presented method has a single oxidation step.
