BACKGROUND: Cell cycle pathway performs an increasingly important character in tumor immunity, presenting unprecedented opportunities for both targeted therapy and immunotherapy for LGG. As a key regulator of mitosis, the mechanisms of MAP7D3 driving malignant process and regulating immune microenvironment of LGG have not been reported.METHODS: This study used multiple bioinformatics analysis methods to analyze the non-negligible role of MAP7D3 in the progression of LGG malignancy by collecting data of 906 LGG patients from TCGA and CGGA databases.RESULTS: Results showed that, firstly, high expressed MAP7D3 in LGG was positively correlated with the malignant features of LGG and negatively correlated with patient overall survival. Simultaneously, multivariate analysis confirmed MAP7D3 as an independent risk prognostic factor for LGG. Secondly, GSEA result proved that MAP7D3 may contribute to the malignant progression of LGG by regulating focal adhesion signaling pathway and also participate in immune-related pathways, such as DNA sensing pathway, B cell receptor signaling pathway, and Toll like receptor signaling pathway. At last, high expression of MAP7D3 in LGG could significantly increased the infiltration of six immune cells and the expression of well-known immune checkpoint PD-1/PD-L1.CONCLUSIONS: Taken together, this groundbreaking study demonstrated that MAP7D3 is an iconic marker of poor prognosis in LGG and a promising target for combination targeted therapy and immunotherapy to relieve the current predicament of treatment resistance and effectively improve quality of life for LGG patients.
