Nitric oxide (NO) is a crucial physiological molecule that plays a regulatory role in maintaining the postharvest quality of fruits and vegetables. While previous studies have acknowledged the potential of NO in preserving the postharvest quality of bamboo shoots, the precise mechanism behind this regulatory function remains elusive. In this particular investigation, we aimed to elucidate the underlying mechanism by identifying key genes involved in NO-mediated regulation of postharvest senescence in bamboo shoots ( Chimonobambusa quadrangularis ). To achieve this, we integrated physiological data with transcriptome data. Notably, our study employed a 0.25 mmol·L − 1 SNP (NO) soaking treatment, which significantly demonstrated its capability to delay senescence, as well as inhibit browning and lignification in bamboo shoots. Transcriptome analysis revealed a total of 5077 genes exhibiting differential expression between the NO treatment group and the control group. Several of these genes were associated with the biosynthesis of lignin, ethylene, and abscisic acid, all of which were notably suppressed. In addition, the expression of genes linked to reactive oxygen metabolism was significantly altered, resulting in a reduction in reactive oxygen species accumulation. Based on our experimental findings, it is evident that NO treatment effectively retards the postharvest lignification and browning of fresh bamboo shoots. This is achieved through the inhibition of lignin biosynthesis, regulation of related plant hormone synthesis, and reduction of reactive oxygen species accumulation, collectively contributing to the delay of postharvest senescence in fresh bamboo shoots. The implications of our study shed light on the potential application of NO treatment in preserving the quality of perishable produce during postharvest handling.
