Modeling demonstrates quantization of Ras activation in membrane corrals, suggesting a processivity-driven tuning mechanism.
Abstract We implemented a rule-based model of Ras activation to examine how SOS processivity could generate discrete signaling outputs. Simulations show that distinct SOS occupancy states produce quantized RasGTP levels, yielding apparent bimodality in small membrane corrals. Increasing Ras deactivation resolves multimodal peaks, while eliminating processivity abolishes quantization. These results suggest a general processivity-driven mechanism for tuning discrete outputs in confined enzymatic systems.
