Deletion of Fam49b increases Th17 cells and energy expenditure in mice, suggesting a link to obesity.
Description CD4 T cell differentiation is affected by the network of cytokines and transcription factors. Fam49b is identified as new negatively regulator of TCR signaling by binding with active rac-1. Previous study showed that Fam49b is essential for thymic development and survival of naïve T cells in the peripheral. However, the role of Fam49b in CD4 T cell differentiation remains unknown. Therefore, in this study, we investigated the effect of Fam49b on CD4 T cell differentiation in Fam49b-deficient mice. In Fam49b-deficient mice, the proportion of intestinal Th17 cells increased, while the proportion of Tregs decreased. We show that Fam49b-deficient mice have a high level of energy expenditure and are resistance to high fat diet induced obesity. Furthermore, the higher energy expenditure characteristic was also observed in Fam49B-deficient chimera mice. The study reveals that Fam49b influences Th17 cell differentiation and plays a significant role in whole body energy metabolism Topic Categories Lymphocyte Differentiation and Peripheral Maintenance (LYM)
