Analysis developed microsatellite markers in Caulerpa denticulata, indicating importance for conservation and population genetics studies.
SUMMARY Polymorphic simple sequence repeat markers for Caulerpa denticulata were developed. In total, 822 sequence contigs possessing microsatellite motifs were identified from transcriptomic sequence data. PCR amplification primers were designed for 80 selected loci, and 20 simple sequence repeat (SSR) markers were successfully developed. The number of alleles per locus ranged from 2 to 5. Observed and expected heterozygosities ranged from 0.125 to 0.525 and from 0.162 to 0.714, respectively. Polymorphic information content (PIC) analysis indicated that 6 of the markers were highly informative while 12 were moderately informative. The combined probability of identity between siblings (CPI‐sib) for the whole set of 20 SSR markers was sufficiently low (4.58 × 10 −3 , 1.02 × 10 −2 ), confirming the usefulness for fingerprinting analyses of in C. denticulata . These polymorphic markers will be valuable for future studies on the conservation and population genetics of this species.
