Project Red Kite develops a framework for malaria vaccine development in the DRC, indicating a new pathway to sovereignty in healthcare.
The stalling of global malaria eradication efforts is attributed to a "fragility triad": biological resistance (antigenic drift), logistical failure (cold-chain dependence), and economic inelasticity (import reliance). This collection presents Project Red Kite (The Kinshasa Protocol), a unified, falsifiable framework for the sovereign elimination of Plasmodium falciparum in the Democratic Republic of the Congo (DRC). Unlike traditional monovalent interventions, this project deploys a systemic "Tetrad of Sovereignty," integrating four independent but mutually reinforcing breakthroughs: Biological: A trivalent PfRh5-CyRPA-Ripr (RCR) fusion antigen that mechanically occludes the erythrocyte invasion pathway with femtomolar affinity ( K_D approximatively 4.2 X 10⁻¹⁵ M). Engineering: A Solid-Core Nanostructured Lipid Carrier (NLC) formulation that stabilizes self-amplifying RNA (saRNA) at ambient tropical temperatures (58 C Glass Transition), eliminating cold-chain requirements. Safety: An Epitope-Erased antigen design validated via ex vivo placental explant modeling to ensure orthogonality to human Basigin (CD147) and reproductive safety. Economic: A Decentralized Manufacturing Model utilizing enzymatic synthesis to achieve a Cost of Goods Sold (COGS) of $1.42 per dose, representing an 84% reduction compared to imported standards. Methodology: This framework was validated using Mamba- State-Space Modeling to generate "Digital Twins" of the molecular, logistical, and immunological systems. By subjecting the candidate vaccine (OMEGA-VAX-001) to >10^6 cycles of in silico falsification before physical synthesis, we present a de-risked "Construction Blueprint" for immediate Phase 1 clinical trials. Artifacts Included in this Collection: Node 1 (Biology): Steric Occlusion of the P. falciparum Basigin-Binding Interface.
